Current Work:
Engineering of Candida tropicalis for ω-hydroxylation of fatty acids (DARPA project: Bioengineering fuel-latent plastics and their conversion to biodiesel).
Long-chain ω-hydroxyfatty acids are used as the monomers that mimic the structure of biodiesel to synthesize a new family of fuel-latent plastics. The plastics will have properties similar to polyethylene but can be degraded by enzyme. After their use they will be shredded and efficiently converted by a mild enzymatic process to provide liquid biodiesel.
In collaboration with DNA 2.0, our lab is developing a whole cell biotransformation process for the conversion of long chain saturated fatty acids to ω-hydroxyfatty acids efficiently and specifically, including the construction of engineered Candida tropicalis using protein engineering (DNA 2.0), screening/testing of the engineered P450 variants, and optimization and process control of fermentation, to obtain the high yield and productivity of ω-hydroxyfatty acids.
